Pramoxine hydrochloride is used as a topical anesthetic to relieve mild pain and itching. The drug was discovered in 1953 at Abbott Laboratories. The trade name of pramoxine hydrochloride is Pramocaine.

Structurally, it is the esther hydrochloride of p-n butoxyphenyl gammamorpholinopropane. The substance stood out as a good topical local anesthetic agent when Abbott Laboratories were researching a series of alkoxy aryl alkamine ethers for such an application. Pramocaine is tolerable by most mucus membranes and ranks low on the sensitizing index, making it unlikely to trigger an allergic reaction.

Pramoxine Hydrochloride is the hydrochloride salt form of pramoxine, a morpholine derivative with local anesthetic property.Pramocaine hydrochloride decreases the permeability of the neuronal membrane to sodium ions by reversibly binding to and inhibiting voltage-gated sodium channels. This results in stabilization of the membrane and, thereby inhibiting the ionic fluxes required for membrane depolarization, hence resulting in the failure to initiate a propagated action potential and subsequent conduction blockade.

Pramoxine is used to temporarily relieve itching and pain caused by minor skin irritation such as minor burns/cuts/scrapes, sunburn, eczema, insect bites, cold sores, or rashes from poison ivy, poison oak, or poison sumac. Some products containing pramoxine are also used to temporarily relieve the itching and discomfort from hemorrhoids and certain other problems of the genital/anal area (such as anal fissures, itching around the vagina/rectum).

Pramoxine hydrochloride cream USP can be used by adults and adolescents as needed three to four times per day. The same dosing instructions can also be followed in the use of pramoxine hydrochloride lotion, pramoxine hydrochloride and menthol gel as well as pramoxine hydrochloride and menthol lotion. Proper dosing for pediatric use in children younger than two years old has not been established. Pramoxine should not be applied across large areas of the body and should not be used for extended periods of time as this may increase the risk of toxicity and systemic absorption.
